INTRODUCTION to diosmin and hessperidin combination

Diosmin and hesperidin is a great combination for vein disorders, such as hemorrhoids, varicose veins, etc. Diosmin has the necessary benefits from the combination, but it works much better together with hesperidin. Moreover, diosmin/hesperidin with the proportion 9:1 or 90:10 are the best formula after numerous experiments and proofs, some other formulas also include Horse chestnut (Aesculus hippocastanum) seed extract, Butcher's Broom Extract (Ruscus aculeatus) (rhizome) (10% ruscogenins), Gotu kola (Centella asiatica) whole plant extract (min 40% asiaticosides),etc.  All these ingredients except diosmin and hesperidin are to achieve the best wanted effects. The most common specification for this combination from our clients is diosmin 90% and hesperidin 10%.

Relations between disomin and hesperidin

Diosmin in is a naturally occurring fiavonoid glycoside that can be isolated from various plant sources or derived from the flavonoid hesperidin. Diosmin was first isolated in 1925 from Scrophularia nodosa, and first introduced as a therapeutic agent in 1969. Diosmin is considered to be a vascular-protecting agent used to help improving chronic venous insufficiency (CVI), haemorrhoids, lymphedema, and varicose veins. As a flavonoid, diosmin also exhibits anti-inflammatory, free- radical scavenging, and antimutagenic properties.
Diosmin differs molecularly from hesperidin by the presence of a double bond between two carbon atoms in diosmin's central carbon ring. Diosmin can be manufactured by extracting hesperidin from citrus rinds, followed by conversion of hesperidin to diosmin, as it is shown below.
Researchers believe that hesperidin and diosmin interfere with arachidonic acid metabolism and histamine release. The flavonoids also inhibit thromboxane and prostaglandin activation. These actions prevent inflammatory responses from occurring, and some believe that the substance may minimize allergic reactions. Physicians commonly prescribe hesperidin and diosmin for patients who are troubled by hemorrhoids. Patients experience decreased pain, swelling and itching while taking the combination formula.
Diosmin has been used for more than 30 years as a phlebotonic and vascular-protecting agent, and has recently begun to be investigated for other therapeutic purposes, including cancer, premenstrual syndrome, colitis, and diabetes.

Mechanism of Action of Diosmin/Hesperidin combination

Diosmin and hesperidin are flavonoid compounds. When used together, they prolong the vasoconstrictor effect of noradrenaline on the venous wall, thus increasing venous return and reducing venous hyperpressure present in patients with chronic venous disease. They also improve lymphatic drainage and protect the microcirculation from inflammation. Diosmin and hesperidin are used together in the treatment of venous disease e.g. chronic venous disease and haemorrhoidal disease.
Diosmin’s mechanisms of action include improvement of venous tone, increased lymphatic drainage, protection of capillary bed microcirculation, inhibition of inflammatory reactions, and reduced capillary permeability. Certain flavonoids, including diosmin, are potent inhibitors of prostaglandin E2(PGE2) and thromboxane A2(TxA2) as well as being inhibitors of leukocyte activation, migration, and adhesion. Diosmin causes a significant decrease in plasma levels of endothelial adhesion molecules and reduces neutrophil activation, thus providing protection against microcirculatory damage.
Inflammatory reactions are trigged by chemical and biological mediators, such as arachidonic acid derivatives (prostaglandins, leukotrienes, or tromboxanes), vasoactive amines (histamine or serotonin), and oxygen free radicals (superoxide ion, 02~, or hydrogen peroxide). In venous inflammation, histamine causes vasodilatation; and PGE2, histamine, and free radicals increase membrane permeability. Diosmin acts by inhibiting the enzyme phosphodiesterase, increasing intracellular cyclic adenosine monophosphate (cAMP) and consequently reducing the level of the main biochemical mediators of inflammation prostaglandin E2 and F2 (PGE2, PGF2), thromboxane B2 (TXB2), and oxygen free radicals"
Diosmin reinforces venous tone by prolonging the activity of pariental norepinephrine. In experiments on the saphenous vein strips of dog, conducted by Heusser & Osswald, diosmin blocked the inactivation of exogenous noradrenaline and caused a slow and gradual contractile response of an oil-immersed strip, which was not attributable to the release of noradrenaline2’. Diosmin exerts a significant potentiation toward NE in both normal and varicose veins under acidotic conditions. Local acidosis depresses reactivity of vascular smooth muscle, especially the response of human isolated saphenous veins to exogenous 14 norepinephrine. In a in vitro study, Juteau and colleagues used isolated varicose veins to test the effect of diosmin+hesperidin and norepinephrine on human rings of veins under acidosis conditions. Results showed that the diosmin+hesperidin combination induced a shift to the left of the concentration-response curves for norepinephrine. This potentiation was significant in both normal and varicose veins and was increased in proportion with the pathological status of the venous rings".
Diosmin enhances the sympathetic activity by increasing nor-adrenaline levels in the synaptic clefts of the vascular wall, reduces the micro vascular high permeability, improves cutaneous micro-circulation as it, normalizes capillary resistance and permeability, inhibits the release of the main pericapillary mediators of inflammation.

The main benefits of Diosmin/Hesperidin 9:1

Diosmin/Hesperidin is the perfect combination for healthy leg veins. This unique natural formula combines two powerful antioxidant flavonoids to promote healthy blood flow in the legs and throughout the body. This combination is good to the veins of the legs. Veins in legs are a problem for many middle or elder age men.

Diosmin/Hesperidin and hemorrhoids

The main function of the disomin and hesperidin is to cure hemorrhoids.  Hemorrhoids appear in about half of all adults by age 50, according to the National Institutes of Health. In the United States alone, that statistic translates into roughly 10 million hemorrhoid sufferers, with about 1 million new cases diagnosed each year.
Hemorrhoids (sometimes called piles) are actually varicose veins located near the rectum or anus. Sometimes the distended veinous tissue protrudes beyond the anal opening; other times the swollen vein remains concealed within the rectum, making its presence known only when symptoms such as pain or bleeding develop.
Diosmin is thought to help reduce inflammation and to increase vein tonicity, two important factors that contribute to hemorrhoids. And taken at larger-than-usual "crisis dose" levels, diosmin also appears to significantly shorten the duration of hemorrhoid bleeding.  One dramatic 2000 Italian study of 66 hemorrhoid patients reported that diosmin decreased pain by 79% and bleeding by 67% during the first week of treatment, followed by an astonishing 98% and 86% reduction in these symptoms by the second week

Diosmin/Hesperidin and varicose veins

Varicose veins and spider veins can be simply a cosmetic concern but usualy varicose veins causes a constellation of symptoms including aching, heaviness of the legs, pain anddiscomfort, particularly after standing up for a long time. Varicose veins can also cause ankle swelling in a few people and sometimes the condition leads to more serious problems. In the United States of America, varicose veins affect as many as 33% of adult women and20% of men.
Flavonoids of diosmin and hesperidin have been shown to strengthen venous tone and improve the elasticity of veins in humans; and to reduce platelet aggregation (clotting) and fibrinogen binding in rats .
Diosmin and hesperidin produced a 200% increase in lymphatic flow, along with decreased capillary permeability and increased capillary resistance, in a study involving dogs. Flavonoids' observed clinical efficacy on edema is believed to be due to improving lymphatic function and reduction of certain adhesion molecules, counteracting chronic venous insufficiency-induced damage to microcirculation. Diosmin may also help the body repair small blood vessel damage that leads to fluid permeability.
